Equipment

  • C18 Sep-Pack cartridge
  • 1 mL Syringe
  • 10 mL Syringe
  • 10 mL Graduated cylinder
  • 100 mL of 18% Isopropanol
  • 10 mL Unsweetened Kool-Aid
  • Distilled water

Procedure

  1.  Prepare the Kool-Aid solution as stated on the package. Do not add any sweeteners.
  2.  Use the 10 mL syringe to flush the C18 Sep-Pack cartridge with 10 mL of 18% isopropanol and time the rate.
  3.  Collect the eluate in a 10 mL graduated cylinder to determine the rate of flow.
  4.  Slowly add 1 mL of the Kool-Aid to the Sep-Pack cartridge.
  5.  Collect the effluent that washes out of the sample as it is being added to the Sep-Pack cartridge.
  6.  Fill the solvent pump with 18% isopropanol solution.
  7.  Pump the solvent through the Sep-Pack cartridge slowly. If the rate is too high, the components of the mixture will not separate well enough. Collect the eluent in a 10 mL graduated cylinder.
  8.  Record the volume of liquid in the graduated cylinder when the red or blue dye is first observed and when the color is no longer observed.
